In this work, we identify CK1α as a novel positive regulator of NOTCH1 signaling and uncover a previously unknown phosphorylation-dependent regulatory mechanism centered on N1ICD residue S1970. By combining cell biology, structural modeling, NMR, mass spectrometry, and Xenopus in vivo assays, we show how CK1α activity supports assembly of the active NOTCH1 transcriptional complex.
