Ammonia
released by growing colonies is absorbed into acidic traps (CitA, 10% citric
acid) in particular intervals of colony development. Nitrogen content in
CitA traps is measured using the Nessler reagent.
Ammonia
is generated in a small vessel containing NH4Cl (70 mg) and 1 M
NaOH (70 ml). The vessel is placed on the plate as indicated on the figure,
in particular times of colony development.
